Comparative transcriptional profiling of the bacterial stress response in temperature and chemically-induced recombinant E. coli processes

چکیده
The expression profiles of selected genes including the product gene, anabolic and stress responsive genes were quantitatively analyzed in cells producing the human basic fibroblast growth factor (hFGF-2), a protein that partially aggregates into inclusion bodies. Transcriptome profiles during temperatureand IPTG-induced synthesis of hFGF-2 using the K12 strain TG1 and BL21(DE3) as production hosts, respectively, were compared.

Expression, purification, and immunization of a chimeric protein containing immunogenic regions of flagellin and intimin proteins against E. coli O157: H7
Introduction: Enterohemorrhagic Escherichia coli (EHEC) and serotype O157: H7 is one of the most important diseases causing diarrhea. Shiga-like toxin secreted by the bacteria destroys epithelial cells and, in acute cases, causes hemolytic uremic syndrome (HUS). Antibiotic therapy is not effective against this pathogen, because it increases the production of Shiga toxin. Designing chimeric immu...

Background: Degradation of phytic acid to inorganic phosphate in domestic animals’ diets requires thermostable phytase. Although Basillus subtilis phytase shows a potential to be degraded phytate complex in high temperature, the enzyme activities and yields need to be increased to make them possible for industrial application. Methods: The phytase gene from Bacillus subtilis DR8886 was...
متن کاملEnhanced Expression of Recombinant Activin A in Escherichia coli by Optimization of Induction Parameters
Activin A is a member of the transforming growth factor β super family. Because of its extensive clinical usages, its recombinant production is beneficial. In this study, activin A was expressed in E. coli using the pET 21a expression vector. The optimization of the activin A production in E. coli was done by using the response surface methodology (RSM). At this stage, the effect of IPTG and la...
متن کاملThe Effect of Heat Shock on Production of Recombinant Human Interferon Alpha 2a (rhIFN α -2a) by Escherichia coli
Recombinant human interferon alpha 2a (rhIFN α -2a) production and cell growth were monitored in a set of genetically modified E. coli strains (MSD1519, MSD1520, MSD 1521, MSD 1522, MSD 1523) producing rhIFN α -2a. The growth was followed at OD 600 nm, changes in cell physiology were detected by pyrolysis mass spectrometry (PyMS) of cell biomass and recombinant protein production was determined...
متن کاملTranscriptional profiling of the bovine hepatic response to experimentally induced E. coli mastitis.
The mammalian liver works to keep the body in a state of homeostasis and plays an important role in systemic acute phase response to infections. In this study we investigated the bovine hepatic acute phase response at the gene transcription level in dairy cows with experimentally Escherichia coli-induced mastitis. At time = 0, each of 16 periparturient dairy cows received 20-40 colony-forming u...
